An efficient two-step methodology of construction of functionalized nucleic acids was developed by a chemo-enzymatic approach using aqueous-phase cross-coupling reactions of nucleotides followed by incorporation by DNA polymerase. The methods are applied in the synthesis of redox-labelled oligonucleotide or DNA probes for electrochemical detection and applications in diagnostics, as well as in the synthesis of modified DNA for applications in chemical biology (regulation of protein binding or bioconjugations).
